What it is

Epitalon (Ala-Glu-Asp-Gly) is a synthetic tetrapeptide derived from the amino acid composition of Epithalamin, a bovine pineal gland extract, and subsequently identified within the pineal gland polypeptide complex itself (PMID 29124531; PMID 40141333). Its proposed mechanisms of action include: (1) Telomerase activation and telomere elongation — Epitalon induces expression of the telomerase catalytic subunit (hTERT), increases telomerase enzyme activity, and extends telomere length in normal human somatic cells and cancer cell lines; in cancer cells, Alternative Lengthening of Telomeres (ALT) activation also contributes (PMID 40908429; PMID 12937682; PMID 15455129). (2) Epigenetic/chromatin remodeling — Epitalon binds preferentially to histone H1/3 and H1/6 and interacts specifically with DNA at CNG and CAG nucleotide sequences, potentially regulating gene transcription via an epigenetic mechanism; it induces deheterochromatinization (decondensation) of heterochromatin in lymphocytes from aged individuals (PMID 32019204; PMID 22117547; PMID 14647006; PMID 24323970; PMID 27259496; PMID 37042594). (3) Melatonin synthesis regulation — Epitalon stimulates arylalkylamine-N-acetyltransferase (AANAT) and pCREB synthesis in pinealocyte culture, increasing melatonin levels; in elderly humans and old monkeys, it restores nocturnal melatonin secretion and circadian rhythm amplitude (PMID 22816096; PMID 17969590; PMID 23289226). (4) Antioxidant effects — Epitalon possesses antioxidant properties, modulates reactive oxygen species (ROS), stimulates antioxidant enzymes (SOD, glutathione peroxidase), and reduces lipid peroxidation products; these effects may exceed those of melatonin in some contexts (PMID 17317455; PMID 60; PMID 18546826; PMID 38). (5) Immune modulation — Epitalon modulates interleukin-2 mRNA levels, influences mitogenic activity of thymocytes, and affects lymphocyte proliferation and cytokine expression (PMID 40141333; PMID 15289226; PMID 35408963; PMID 7). (6) Anti-senescence — Epitalon targets the SUV39H1 axis to partially eliminate therapy-induced senescent cells in NKTCL models. (7) Neurogenic differentiation — Epitalon increases expression of neurogenic differentiation markers (Nestin, GAP43, β-Tubulin III, Doublecortin) in human gingival mesenchymal stem cells. (8) Neuroendocrine effects — Intranasal administration increases cortical neuron discharge frequency in rats. The precise primary receptor or molecular target remains uncertain.
